Item Acute effect of centrally injected Nesfatin-1 on some blood electrolytes and metabolites in rats(Bursa Uludağ Üniversitesi, 2020-11-18) Bayram, Gökçen Güvenç; Ülger, Ebru Yalçın; Yalçın, Murat; Veteriner Fakültesi; Fizyoloji Ana Bilim Dalı; 0000-0002-1413-3651; 0000-0003-1756-1288; 0000-0002-5600-8162Nesfatin-1 is a newly found food and water intake regulatory neuropeptide. Because it can regulate nutrition and thirst, nesfatin-1 may also have the potential to affect levels of blood electrolytes and metabolites. The current study was intended to resolve the acute influence of intra- cerebroventricularly injected nesfatin-1 on the levels of some blood electrolytes and metabolites in rats. The experiments were conducted on Sprague Dawley male rats. Nesfatin-1 (200 pmol) or saline (5 μL) was given the rats intracerebroven- tricularly. Central nesfatin-1 treatment caused increases in the concentrations of blood glucose, lactate, hematocrit, and hemoglobin without changing the blood pH, creatine, Na, K, Ca, Cl, and HCO3 levels. In conclusion, our findings show that the central nesfatin-1 could affect the concentrations of blood glucose, lactate, hematocrit, and hemo- globin without altering the blood electrolytes. This could be interpreted as the secondary effect of nesfatin-1 as a consequence of centrally injected nesfatin-1-evoked activation of sympathetic nerves.Item Alimentary lymphoma in a cat(Bursa Uludağ Üniversitesi, 2018-05-18) Karakuş, Adil Ömer; Kasap, Sevim; Mutlu, Ayşe Meriç; Tavus, Melike Akbala; Salcı, Hakan; Sönmez, Gürsel; Veteriner FakültesiLymphoma is the most common feline malignant tumor, and the alimentary form is the most common seen form. Alimentary form in the cat typically involves lesions of the small intestine, stomach, mesenteric lymph nodes, and liver. The disease is often reported in cats between 1 and 20 (mean 13 years) years. A 3-year-old domestic male cat was admitted to Uludağ University Animal Hospital due to chronic weight loss, loss of appetite, diarrhea and vomiting traced back to 15 days. Abdominal ultrasonography was performed and a large solid mass was spotted in the cranial abdomen. During the treatment, the cat developed respiratory stress, shock and died 2 days after. In light of the clinical symptoms, ultrasonography, necropsy and histopathological findings; small-cell-alimentary lymphoma was diagnosed for the 3-year-old cat. It can be interpreted that small cell (lymphocytic) alimentary lymphoma is seen rarely in younger cats and seldomItem Allometric growth of non-carcass components and hind limb tissue composition in saanen goat kids reared under natural and artificial systems(Bursa Uludağ Üniversitesi, 2022-07-26) Keçici, Pembe Dilara; Yalçıntan, Hülya; Ekiz, BülentThe aim was to investigate the growth of certain non-carcass components and hind limb tissues using allometric coefficients in Saanen goat kids reared under natural and artificial systems and slaughtered at 80 days or 120 days of age. 42 Saanen kids were used in the study [(24 artificial reared kids (SA-80: 12 kids, SA-120: 12 kids), 18 natural reared kids (SA-80: 9 kids, SA-120: 9 kids)] for determination of indicated parameters. The effects of rearing system were not significant in weights of non-carcass components and hind limb tissue compositions, which indicate that, even though they were fed with milk replacers, artificial reared kids can have similar growth performance when they are slaughtered at similar age with natural raised kids. However, slaughter age had significant effect on many non-carcass components, since the weight of the organs increases in parallel with animal’s live weight. It is seen that allometric growth of non-carcass parameters, except skin and testis in both rearing groups and stomach and intestines in natural rearing, was slower (b<1) than EBW of kids slaughtered at 120th days of age. This result shows that when the Saanen kids will be slaughtered at later ages, the increase of live weight will be mainly due to the increased saleable carcass components. Additionally, tissue compositions showed that muscle growth continued rapidly (b=1.494 for natural and b= 1.251 for artificial reared) in both groups on the 120th day of age. This situation reveals that prolonging the fattening period with Saanen kids can increase meat production for both rearing systems. On the other hand, it is observed that the amount of both subcutaneous and intermuscular fat increased more rapidly at the 120th day of age in both groups. Therefore, the optimal slaughter age at 120th days should be determined by taking into account the possible increasing amount of fat.Item Alterations in some blood parameters after high level ethanol ıntake(Uludağ Üniversitesi, 2005-06-07) Yağcı, Artay; Yalçın, Murat; Veteriner FakültesiThe effects of high level ethanol intake on some blood parameters over a 50 days period were examined in 36 Sprague Dawley rats divided into 3 treatment groups: Ad libitum fed control group (n=12), sucrose group (n=12) fed with solution of sucrose so calorie intake equated with the ethanol group, and ethanol group (n=12) added ethanol (15 % v/v) in drinking water. Significant decreases (p<0.05) were noted in ethanol group’s white- and red-blood cell counts, haemoglobin concentration, erythrocyte diameter, erythrocyte sodium and potassium levels, while significant (p<0.05) increases were observed in their mean corpuscular volume and mean corpuscular haemoglobin levels. No changes were observed in hematocrit level. Results of this study suggest that high level ethanol intake in rats causes alterations in blood cells count and in erythrocyte diameter and in erythrocyte element composition and this negative effect of ethanol on blood parameters did not stem from the energy of ethanol.Item Analysis of IGf1 C472t and TG C422t polymorphisms in Turkish grey steppe and holstein crossbred bulls(Uludağ Üniversitesi, 2018-03-01) Ardıçlı, SenaThe aim of the study was to determine genotypic distribution of bovine insulin-like growth factor 1 and thyroglobulin genes in Turkish Grey Steppe (Boz) and Holstein crossbred bulls. In the study, unrelated 30 Turkish Grey and 26 Holstein crossbred cattle were used. Genomic DNA was isolated from whole blood using standart phenol-chloroform extraction method. The polymerase chain reaction-restriction fragment length polymorphism technique was used for genotyping of C472T and C422T polymorphisms in insulin-like growth factor 1 and thyroglobulin genes, respectively. Hardy-Weinberg equilibrium was calculated by using PopGene software program. In addition the population indexes including heterozygosity, number of effective alleles and polymorphism information content were determined on the basis of allelic distribution. For insulin-like growth factor 1 gene, A allele frequency was higher and BB genotype was not present in both breed groups. For thyroglobulin gene, TT genotype was not present and moreover C allele was almost fixed in both Turkish Grey and Holstein crossbred bulls. In addition, extremely low minor allele frequencies in resulted in low genetic variabilities of population indexes. The present results may be useful for future studies conducted on bovine genome with respect to insulin-like growth factor 1 and thyroglobulin genes.Item Animal originated allergens and measures to reduce exposure(Uludağ Üniversitesi, 2009-12-16) Coşkun, Şevki; Veteriner FakültesiMany of the animal allergens widespread in the world sources from mammalian or arthropod animals. Sinem Özdemir; İpek, Volkan; Cangül, İ. Taci; İntaş, Kamil Seyrek; Veteriner FakültesiA 3.5-year-old, pregnant Pekingese was presented with signs of dystocia. Vaginal examination revealed no fetuses in the birth canal, and no heart beats were observed in the fetuses, ultrasonographically. Four dead fetuses were retrieved by en bloc ovariohysterectomy. All fetuses had hind limb deformities and intestinal evantration was observed in three fetuses. A diagnosis of arthrogryposis and abdominal wall defect complicated with intestinal evantration was made after the macroscopic examination of the fetuses. The owner did not permit necropsy. To the best of our knowledge, this is the first report of arthrogryposis and abdominal wall defect complicated with intestinal eventration encountered in Pekingese fetusesItem Assesment of some udder and teat traits of honamlı goats in terms of dairy characters(Bursa Uludağ Üniversitesi, 2019-10-20) Akbaş, Aykut Asım; Elmaz, Özkan; Sarı, Mehmet; Saatçi, MustafaStudy was conducted to evaluate the dairy characters of Honamlı goats by using ADGA scoring system. For this aim, eight Honamlı goat flocks reared under local breeder condition in Antalya and Burdur provinces were used. Five goats from each flock totally 40 goats with the highest score were defined. Some udder and teat characteristics of these goats were determined during six months (March-August). While Honamlı goats would be given in the acceptable category for dairy character with their numerical final scores (70-79); relatively higher values were detected from the flocks in Antalya province than Burdur ones (P<0.05). The udder characteristics increased in April based on course of lactation of Honamlı goats in the best flocks (Antalya province) were determined as follows, udder depth, udder circumference, udder width, teat length, teat diameter, the distance between teats and the distance to floor from the teat 21.73 cm, 46.97 cm, 17.06 cm, 8.05 cm, 3.08 cm, 14.47 cm and 37.22 cm, respectively. It was thought that results of study were important to be an introduction for following studies that is going to be potential goats having dairy characteristics among Honamlı goatsItem Assessment of microbiological contamination factors in frozen stuffed snail processing stages(Uludağ Üniversitesi, 2006-02-28) Günşen, Uğur; Dokuzlu, Uğur; Temelli, Seran; Şen, M. Cem; Veteriner FakültesiIn this study, six predetermined processing stages and six possible contamination sources in frozen stuffed snail production were examined for the enumeration of total aerobic mesophilic bacteria, coliforms, Escherichia coli (E. coli), coagulase positive staphylococci and for the detection of Listeria spp. and Salmonella spp. Results revealed that the cooling stage was a major contamination factor. Also, shells, although ‘pasteurized’, seemed to be one of the causes for the increases in the final microbial counts of the product. Another minor source for recontamination was determined as the cooling vat. Improvements were recommended for more effective technology applications, which would eliminate risks for contamination for the product.Item Bir atta karşılaştığımız pododermatitis chronica verrucosa (Ayak Kanseri) olgusu'nun operatif sağıtımı(Uludağ Üniversitesi, 1984) Görgül, O. Sacit; Veteriner FakültesiThe case of the Chronic Proliferative Pododermatitis (Foot Canker) had been occured in an English thoroughbred mare. The lesions of the Canker were found both of the hind limbs. Central and lateral frog clefts and soft hell bulbs were affected. Operation was made under general anaesthesia by using intraveneous 10 % Chloral hydrate solution. Affected Horn parts were totally resected. After the operation an antiseptic compress was applied by using 5 % Sulfate de Zinc solution. Then the bandage was changed and a Penicillin paste was applied. Following every 10 days the bandage was changed. The surgical repair was succesfuly resulted after two mounts.Item Azerbaycan propolislerinin fenolik madde içerikleri ve kalite açısından değerlendirilmesi(Bursa Uludağ Üniversitesi, 2019-09-14) Sariyev, Rashad; Çayci, Meltem; Oruç, Hasan Hüseyin; Veteriner Fakültesi; 0000-0002-1129-4118; 0000-0002-3401-9186; 0000-0002-5399-2395Propolisin antioksidan, antimikrobiyal, immunostimülan ve antitümöral gibi biyolojik etkileri, içeriğindeki fenolik bileşiklerden kaynaklanır. Fenolik bileşiklerin nitel ve nicel analizleri propolisin kalitesini değerlendirmede çok önemlidir. Bu çalışmanın amacı, Azerbaycan’dan toplanan ham propolislerde, faydalı biyolojik özellikleri bilinen fenolik bileşiklerin nitel ve nicel olarak analizini yapmak ve sonuçları değerlendirmektir. Propolis örnekleri Azerbaycan’ın 5 ilinde bulunan, toplam 14 farklı arılıktan, kovanın farklı kısımlarından kazınarak, 2018 sonbahar mevsiminde toplandı. Toplam 14 ham propolis örneğinde, 17 hedef fenolik bileşiğin (flavonoid ve fenolik asitler) HPLC-DAD sistemiyle analizi yapıldı. Analizi yapılan tüm örneklerde epigallokateşin gallat, kafeik asit, ferulik asit ve isoferulik asit tespit edildi. Propolisin toplandığı şehirlere göre propolis örnekleri arasında fenolik bileşikler ve miktarları bakından farklılıklar olduğu görüldü. Bu çalışmada Azerbaycan propolisinin kalitesinin genel olarak iyi olduğu ve fenolik madde içeriğinin Avrupa ve Türkiye’deki propolislere benzer olduğu tespit edilmiştir.Item Biochemical factors ınfluencing autolysis of leuconostocs in buffer(Uludağ Üniversitesi, 2010-07-12) Çıbık, Recep; Veteriner FakültesiFactors influencing autolysis of two dairy leuconostocs strains chosen for their moderate and relatively higher autolytic extend were investigated in buffer system. Higher level of autolysis was observed in bacteria that were harvested at earlier exponential phase. Autolysis was greatly influenced in acidic and basic pH values and optimal level was monitored between 6 and 7 values. Optimal incubation temperature to induce lysis was detected at 30°C, which is at the same time optimal growth temperature for leuconostocs strains. Whereas autolysis was activated by the presence of monovalent cations, an important level of inactivation was noted in the presence of divalent cations.Item Biomechanical comparison of the effects of the storage temperature on Tibiotarsus in Japanese quail(Bursa Uludağ Üniversitesi, 2021-12-18) Süzer, Bayram; Veteriner Fakültesi; Anatomi Ana Bilim Dalı; 0000-0002-2687-1221The study aimed to compare the effects of different cryopreservation temperatures on mechanical properties and determine the optimal cryopreservation temperature for bones in Japanese quail. Bone biomechanical tests are getting more attention but, fresh bones are not always available for testing and have a limited lifespan. Cryopreservation of biological specimens is often needed during tissue preparation and me- chanical testing. In the study, the tibiotarsi were collected from 8 weeks of age quail, and bones were divided into four groups of fresh bones; frozen at 0 o C, frozen at -20 o C, and frozen at -80 o C. Frozen bones were kept in the freezer for three weeks. After three weeks, bones were subjected to a three-point bending test for biomechanical evaluation. There was no significant difference between the mechanical strength properties of fresh tibiotarsi and the tibiotarsi stored in three different storage conditions of 0 o C, -20 o C, or -80 o C. It was observed that cryopreservation of tibiotarsi at 0, -20, and -80 °C for up to three weeks did not negatively affect bone biomechanical properties in quail.Item Bovine viral diarrhea virus propagates in cell culture after ınoculation of ınactivated sera from persistently ınfected calves(Uludağ Üniversitesi, 2016-12-12) Göktuna, Pelin Tuncer; Yeşilbağ, Kadir; Veteriner FakültesiDuring routine diagnostic investigations on submitted serum and swab samples from three (n:3) calves in our laboratory for detecting BVDV antibody titers and antigen we noticed that successful virus propagation can occur from heat inactivated (in a water bath at 56° C for 30 min) 2 serum samples. For further confirmation of virus propagation in heat inactivated serum samples, 11 sera which were previously detected positive for BVDV were also investigated. As a result, from a total of 14 heat inactivated-field originated sera samples virus propagation was confirmed in 4 (28.5%) samples. This result suggests that heat inactivation of the serum is not eliminating the risk of pestivirus contamination, thus interfere the results of virus isolation and serological diagnostic methods including serum neutralization. Moreover relative resistance of BVDV to heat inactivation may lead to failure in complete inactivation of the virus in fetal calf sera which is an important contaminant for biologicals like cell cultures and attenuated live vaccines.Item The case of gastrointestinal nematodiosis forming a nodule in a gazelle’s (gazella subgutturosa) abomasum(Bursa Uludağ Üniversitesi, 2021-05-05) Yılmaz, RahşanThis study has reported the case of abomasal teladorsagiosis in gazelle (Gazella subgutturosa) that was found dead while under care at a wildlife reserve area in Sanliurfa, Turkey. Necropsy was performed on six-year-old, male gazelle, which was found dead in its natural environment and brought to the Department of Pathology, Faculty of Veterinary Medicine, Harran University. At necropsy were observed became whitish in color and thickened the plicae spirals on the abomasum mucosa. In the microscopic examination, parasites were encountered on the mucosa abo masum and lumen of the glands in lamina propria. Also in the lumen of the glands with parasited were detected necrotic debris and neutrophil leukocyte accumulation and to increase in the connective tissue between the glands. In the scannig electron microscopic examination, parasites were observed on the abomasum mucosa. This study is the first report case describing gastrointestinal nematodiosis forming a nodule in the abomasum of in a gazelle in Turkey.Item Changes in serum protein and lipoprotein fractions during various lactation stages and dry period in dairy cows(Uludağ Üniversitesi, 2012-01-25) Güzel, Saime; Tanrıverdi, Meltem; Polat, Ümit; Veteriner FakültesiThe aim of this study was to determine the changes in the serum albumin, alpha 1 (α1)-globulin, alpha 2 (α2)-globulin, beta (β)-globulin, gamma (γ)-globulin, and alpha-lipoprotein (high density lipoprotein-HDL), pre-beta-lipoprotein (very low density lipoprotein-VLDL) and beta-lipoprotein (low density lipoprotein-LDL) and total lipid fractions of the cows in dry period and various stages of lactation period. Fourty healthy, four years old, Holstein dairy cows were used as material. Ten of them were in dry period and the rest were in lactation period. Between the days 48-51, 147-161. and 180. and over of lactation period were accepted as first, second and third stage, respectively. There were 10 cows in each lactation stages. Serum protein and lipoprotein fractions were compared between the dry period, first, second and third stages of lactation period. While significant differences were determined for α1-globulin, β-globulin, γ-globulin, HDL, VLDL, LDL and total lipid levels, no significant differences were observed in albumin and α2-globulin between dry period, first, second, third stages of lactation periodItem Changes in the gene expression of pyruvate dehydrogenase kinase isoenzymes during early differentiation of mouse embryonic stem cells(Bursa Uludağ Üniversitesi, 2022-09-12) Güzel, Saime; 0000-0003-0796-5000The embryonic stem cells (ESCs) are pluripotent, self-renewing cells that are able to differentiate into any of the germ layers involved in embryogenesis. However, the molecular mechanisms that control ESC pluripotency and differentiation remain poorly understood. The family of Pyruvate dehydrogenase kinase (PDK1-4), inactivates the mitochondrial pyruvate dehydrogenase complex via phosphorylation, plays a crucial role in the control of glucose homeostasis. In the current study, gene expression levels of PDK isoenzymes were analyzed on undifferentiated mouse embryonic stem cells (mESCs) and compared to mESCs induced to differentiate by removal of leukemia inhibitory factor (LIF) for 1, 3 and 5 days. Besides, gene expression analysis of several genes related to pluripotency and differentiation were performed by real-time quantitative PCR. In addition, glucose uptake rates in early differentiated and undifferentiated mESCs were determined using a colorimetric assay kit. Differently expression level of PDK isoenzymes in pluripotent and LIF-depleted mESCs suggest that they may have roles in differentiation and pluripotency of ESCs. Furthermore, this study lays the foundation for detailed investigation of molecular mechanisms underlying the roles of PDKs in the pluripotency and transition to differentiated state of ESCs.Publication Characterization and computational ınvestigation of polymorphisms in the TIR domain of the TLR4 protein in holsteins raised in Turkey(Bursa Uludağ Üniversitesi, 2023-03-01) Atalay, SertaçIncreasing environmental temperatures due to climate change adversely affect livestock welfare and health. Moreover, temperatures increase thedistribution and survival of parasites and infectious agents. Livestock diseases that cause significant economic losses are a worldwide concern.Toll-like receptor 4 (TLR4) is an ideal marker gene candidate, due to its critical role it plays in initiating the immune response against pathogens.In this study, the toll-interleukin-1 receptor (TIR) domain polymorphisms were investigated in the Holsteins raised in Turkey. by DNAsequencing. The effects of polymorphisms on the protein structure and function were evaluated by computational tools (I-Mutant Suite, ProjectHope and PyMOL). The rs8193069 C>T polymorphism was detected in the TIR domain. The SNP causes Threonine to Isoleucine substitutionat position 674 of the TLR4 protein which is the second amino acid of the TIR domain. The I-Mutant Suite predicted that Thr674Ile substitutioncould decrease protein stability (DDG= -0.40 Kcal/mol). Project Hope results showed that mutant and wild-type amino acids have differentproperties and that this can disturb the TIR domain. The Thr674Ile polymorphism observed in the TIR domain of TLR4 protein in Holsteinsraised in Turkey might affect the function of the TLR4 by causing physicochemical changes.Item Clinical and biochemical evaluation of cows occurring severe weight loss after calving(Uludağ Üniversitesi, 2010-04-05) Şentürk, Sezgin; Mecitoğlu, Zafer; Temizel, Mutlu; Cihan, Hüseyin; Kasap, Sevim; Demir, Gülşah; Veteriner FakültesiThis study was designed to observe the biochemical and clinical changes in Holstein cows suffering from severe weight loss after calving. Clinically, out of the 75 milking cows in the first 8 week postpartum examined, 25 cows aged 4– 8 years were found to be suffering from decreased milk production, severe body weight loss. 25 healthy cows in the same period after calving, aged 4 – 8 years, were also used as a control group. Serum biochemistry analyses including potassium (K), sodium ( Na), calcium (Ca), phosphorus (P), magnesium (mg), albumin, alkaline phosphatase (ALP), gamma-glutamyltransferase (GGT), aspartate aminotransferase (AST), total biluribin, (TBIL) total protein (TP), creatinine (Cre), blood urea nitrogen (BUN), and β-Hydroxybutyrate (BHBA) were evaluated. Body condition score (BCS) of 17 of 25 cows, severe weight loss after calving cows, was ≤ 2.0 and BCS of 8 cows was between 2.0 and 3.0. BCS of Animals in the control group were determined as range: 3.5 -4.0. Endometritis in 11 cows, subclinical mastitis in 8 cows, clinical ketosis in 5 cows, sub clinical ketosis in 20 cows, and left displaced abomasum in 3 cows were determined. Identical points of 25 animals were higher BHB, GGT, AST, total globulin, and total protein levels according to healthy cows and references values. In addition, these cows had hypoalbunemi and hypocalcaemia, when compared with healthy cows. The results of the study reported here indicated that metabolic profile including serum BHBA, calcium, albumin, GGT, AST, total protein, should be evaluated to determine the causes of severe weight loss, and decreased yield in cows after calving.Publication Comparison of beta-hydroxybutyrate levels in the first three weeks after calving in four healthy different breeds of cows(Bursa Uludağ Üniversitesi, 2023-01-18) CİHAN, HÜSEYİN; KASAP, SEVİM; ŞENTÜRK, SEZGİN; Veteriner Fakültesi; İç Hastalıkları Ana Bilim Dalı; 0000-0003-1544-6438; 0000-0001-9501-9983; 0000-0002-2465-9913The objective of this study was to determine the differences between the healthy Holsteın-Friesian, Jersey, Simmental, and Aberdeen Angusbreeds Beta-hydroxybutyrate (BHBA) levels at the first three weeks after calving. The blood samples were collected for analysis between 10-20 days postpartum. In this study, 80 animals were used in different regions in total. BHBA was analyzed by Ketosite® (Ketosite®, Stanbio LabrotoryInc., Boerne, TX USA). Milk fat and protein levels were immediately measured by an electronic milk analyzer (Milkoscan FT1 FOSS Industries, Denmark). Statistical analyses of the study were performed using Sigma Plot 12 (Systat Software Inc., San Lose, CA). The means of BHBA levelswere compared by One Way Anova. As in Holstein cows, blood BHBA levels increased in the early lactation period and also in other breeds. Theresults of this study presented that serum BHBA levels were significantly higher in Holsteın-Friestein cows than in Simmental and AberdeenAngus cows.
