Item Acute effect of centrally injected Nesfatin-1 on some blood electrolytes and metabolites in rats(Bursa Uludağ Üniversitesi, 2020-11-18) Bayram, Gökçen Güvenç; Ülger, Ebru Yalçın; Yalçın, Murat; Veteriner Fakültesi; Fizyoloji Ana Bilim Dalı; 0000-0002-1413-3651; 0000-0003-1756-1288; 0000-0002-5600-8162Nesfatin-1 is a newly found food and water intake regulatory neuropeptide. Because it can regulate nutrition and thirst, nesfatin-1 may also have the potential to affect levels of blood electrolytes and metabolites. The current study was intended to resolve the acute influence of intra- cerebroventricularly injected nesfatin-1 on the levels of some blood electrolytes and metabolites in rats. The experiments were conducted on Sprague Dawley male rats. Nesfatin-1 (200 pmol) or saline (5 μL) was given the rats intracerebroven- tricularly. Central nesfatin-1 treatment caused increases in the concentrations of blood glucose, lactate, hematocrit, and hemoglobin without changing the blood pH, creatine, Na, K, Ca, Cl, and HCO3 levels. In conclusion, our findings show that the central nesfatin-1 could affect the concentrations of blood glucose, lactate, hematocrit, and hemo- globin without altering the blood electrolytes. This could be interpreted as the secondary effect of nesfatin-1 as a consequence of centrally injected nesfatin-1-evoked activation of sympathetic nerves.Item Determination of opportunistic pathogens and antimicrobial resistance characterization Isolated from Rainbow trout in Turkey(Bursa Uludağ Üniversitesi, 2020-10-29) Satıcıoğlu, İzzet Burçin; Duman, Muhammed; Özdemir, Burak; Ajmi, Nihed; Altun, Soner; Veteriner Fakültesi; Su Hayvanları Hastalıkları Ana Bilim Dalı; 0000-0001-7707-2705; 0000-0001-9096-875XAquaculture, by modern and technological methods, ensures effectual means for healthy fish production under “controllable” conditions. However, fish have disease problems due to a lack of imbalance with microbiota or opportunistic pathogen in culture systems. In the pres- ent study, we identified the opportunistic pathogens and some members of microbiota, which were obtained from our culture collection by culturable methods and characterized biochemically in addition to molecular analysis. We identified the isolates by partial gene sequencing and confirmed taxonomically in the bacterio.net database. The isolates were characterized based on antimicrobial susceptibilities by broth microdilution method analysis, and the resistance gene determinants were screened by PCR analysis. A total of 14 species were identified with high genetic similarity in the GenBank database. MIC results showed that bacteria have heterogeneous characteristics for the susceptibility of an agent into the genus, and species have high MIC values for sulfamethoxazole, trimethoprim, and ampicillin comparing to other agents. A total of 13 different resistance genes were determined in the bacteria, and some of them have multiple resistance genes up to five genes.Item Easy isolation, propagation, characterization and multilineage differentiation of equine amniotic fluid derived stem cells(Bursa Uludağ Üniversitesi, 2020-10-28) Kibria, Golam; Nawaz, Shah; Akkaya, Özlem Özden; Dikmen, Tayfun; Yağcı, ArtayEasy harvesting and handy application of stem cells therapy want convenient and alternative source. Equine amniotic fluid derived mesenchymal stem cells make hope for scientists as of no ethical concern and of higher proliferation and differentiation potency. Equine AFSCs were isolated, cultured and propagated for characterization of potency of growth and multilineage differentiation study. Successful harvesting and optimum differentiation of equine AFSCs into osteogenic, adipogenic and neurospheres through this study could make a ground for their clinical application for various purposes. Further extensive and comprehensive molecular studies are important to conclude the precious procedures for clinical approach of amniotic fluid derived stem cells.
