Fresh and/or frozen semen was diluted a concentration of 50 x 10° spermatozoa/ml in physiologic saline. An equal volume of se men solution and 5 % gelatin solution in physiologic saline was mixed to prepare antigen. Blood serum and seminal plasma of bulls and blood serums obtained from heifers, pregnant cows and infertile cows and some cervical mucus of cows were used as agglutinin sources. For the test double dilution of the serum samples were made in physiologic saline containing 2.5% ge latin. In plastic hemagglutination wells 50 microlitre of each serum dilution was placed in a clean well and equal volume of sperm antigen (20-30 x 106 spermatozoa/ml) was added to each well and the contents mixed gently. The mixture in each well was allowed to rise and fill in to the capillary tube. The tubes are placed upright in clay and are incubated at room temperature. Results were recorded at 2 hours. The appearance of white floccules along with clearing of the suspending medium was evaluated as a pozitif reaction. The lowest anti-sperm agglutinins were determined in blood serum of pregnant cows and heifers. Agglutinins were detected in some blood serums of bulls and cows. Increased titers of agglutinins (1: 32) were found in infertile cows. Presence of high titer of sperm agglutinins in most of infertile cows might be an explanation of some cases of infertility in cows.
