Araştırma 27’şer merinos ırkı kuzudan oluşan deney ve kontrol grubu olmak üzere toplamda iki gruptan oluşmuştur. Çalışma süresi boyunca her iki gruptaki kuzulara kaba ve konsantre yemler ad libitum olarak verilmiştir. Grupların kuzu besi yemleri içerik ve kompozisyon olarak benzer düzenlenmiş, ek olarak deney grubununkine %0,4 oranında sodyum bütirat katılmıştır. Her 2 gruptaki kuzularda da doğumda, deneme başlangıcında, 45 günlük yaşta, sütten kesim zamanında ve deney sonunda canlı ağırlık tartımları yapılmıştır. Grupların yemliklerine konan ve artan konsantre ve kaba yemler tartılmıştır. Deney sonunda, grupların sıcak karkas ağırlıkları saptanmıştır. Her iki gruptan 3’er kuzu rastgele seçilerek rumenleri alınmış, rumen papilla uzunlukları ve kalınlıkları ölçülmüştür. Bulgularda, deney grubunun toplam canlı ağırlık artışlarının kontrol grubuna göre 45. ve 105. günlerde daha yüksek olduğu görülmüştür (p<0.05). Deney grubunun ortalama sıcak karkas randımanları kontrol grubuna göre daha yüksek olmuştur (p<0.05). Kontrol grubun araştırma sonu ortalama toplam kuru madde tüketimi deney grubuna göre 4,01 kg daha fazla olmuştur. Deney grubunun 1 kg karkas maliyeti kontrol grubuna göre daha düşük olmuştur. Kontrol grubunun rumen papilla uzunlukları ve genişliklerinin deney grubuna göre daha fazla olduğu saptanmıştır. Sonuç olarak, kuzu besi yemlerine katılan %0,4 oranındaki kaplanmış sodyum bütiratın besi performansını olumlu etkileyebileceği, ancak rumen gelişimine üzerindeki etkilerin daha iyi anlaşılabilmesi için sütten kesimden önce de histolojik incelemeler yapılmasının faydalı olacağı kanısı oluşmuştur.Publication Morphometric examination of hind limb and foot bones and fibre type composition of crus region muscles in quail and pigeon(Wiley, 2023-02-12) İşbilir, Fatma; Akkoç, Cansel Güzin Özgüden; Arıcan, İlker; ÖZGÜDEN AKKOÇ, CANSEL GÜZİN; ARICAN, İLKER; Veteriner Fakültesi; Anatomi Ana Bilim Dalı; 0000-0003-0712-0892; 0000-0001-6342-0094; F-2822-2017; AAG-7518-2021In this study, the foot and hind limb bones of pigeons and quails were measured morphometrically. Additionally, microscopic classifications of the muscles affecting the foot and digit joints were made. For the macroscopic inspection, 40 birds were used, including 20 adult quails (10 males, 10 females) and 20 adult pigeons (10 males, 10 females). Diethyl ether was inhaled to anaesthetize the animals. The poultry animals were put under anaesthesia, and radiographic pictures of their left feet were obtained individually. DAP measurements were performed separately from the images taken with the Image J program. Then, they were euthanized by cervical dislocation under diethyl ether anaesthesia. The right legs of the euthanized animals were preserved in a 10% neutral formalin solution for histology procedures just after the legs were dissected from the trunk. Morphometric measurements of bone lengths were made in accordance with the measurement points specified by von den Driesch. After fixation for histological examination, routine tissue follow-up was performed and the tissues were embedded in paraffin. The presence of SO-type I, FG-type IIb and FOG-type IIa in 4-5 mu sections taken from paraffin blocks was demonstrated using the indirect streptavidin-biotin-complex method from immunohistochemical methods. The result of our study was statistically evaluated at p < 0.05 and p < 0.001 levels. The length of the hallux, the articulation point to the TMT and the fibre arrangements in the two flexor group muscles showed that the hind limbs and feet of the pigeons had a more favourable anatomical and histological structure for the perching movement.Publication Immunohistochemical demonstration of mast cells in bovine papillomatous digital dermatitis of holstein-friesian dairy cattle(Hellenic Veterinary Medical Soc, 2023-10-01) Özgüden-Akkoç, C. This study aims to evaluate MCs distribution and heterogeneity in healthy and digital dermatitis lesions of dairy cattle for the first time. A total of 50 skin samples, 25 healthy and 25 with digital dermatitis (DD) lesions were sampled in Holstein-Friesian dairy cattle. All samples were stained with hematoxy-lin-eosin (H&E) for microscopic examination and also stained with Toluidine blue for MCs demonstration. Epidermal acanthosis and hyperkeratosis with dermal inflammatory cell infiltrations were in consistent with digital dermatitis lesions. Spirochetal agents were successfully demonstrated with Warthin-Starry staining in 22 out of 25 DD samples. Increased number of mast cell was observed in digital dermatitis samples when compared with healthy skin samples. The average number of intact MCs was 4.6 +/- 2 and 9.3 +/- 1 in healthy and digital dermatitis samples, respectively. Two-fold increase in the number of intact MCs in digital dermatitis samples was observed. Degranulated mast cell numbers in tissue sections were also higher in digital dermatitis samples and 4.25-fold increase was recorded in affected skin samples. Immunophenotype of MCs in skin samples were identified by immunohistochemical staining with anti-tryptase and chymase antibodies. Only tryptase positive MCs were observed in healthy and DD samples. In the statistical analysis, differences in the mean intact and degranulated MCs were found to be significant (mean intact: p<0.05, mean degranulated: p<0.01). Our results possibly suggest that MCs may have important roles in the pathogenesis of bovine digital dermatitis.
