Abstract
Bu çalışma, California Mastitis Test (CMT), Direkt Mikroskopik Somatik Hücre Sayımı(DMSHS), Ayırıcı hücre sayımı bulgularına dayanarak normal ve subklinik mastitisli olarak ayrılan süt örneklerinde çeşitli biyokimyasal testleri karşılaştırmalı olarak incelemek ve bu testlerin ineklerde subklinik mastitislerin teşhisinde kullanılabilirliğini saptamak için yapıldı. Bu çalışmada, Uludağ Üniversitesi Veteriner Fakültesi Uygulama ve Araştırma Çiftliğinde ve Bursa bölgesindeki ticari sütçü işletmelerde bulunan 3-7 yaşlı, Holstein, Montofon ırklarına ait 100 adet inek araştırma materyali olarak kullanıldı. Bütün ineklere, ilk önce CMT uygulandı. CMT sonuçlarına göre (-) reaksiyon gösteren 30 meme lobu sağlıklı, (?, +,++, +++) reaksiyon gösteren 70 meme lobu subklinik mastitisli olarak ayrıldı. Somatik hücre sayıları (SHS), subklinik mastitisli süt örneklerinde 1.087.571 ±58.489 hüc/ml iken, normal süt örneklerinde 134.666±1 5.798 hüc/ml olarak bulunduCPOjOOI). Ayırıcı hücre sayımı bulguları; toplam SHS'nın, normal süt örneklerinde %83,41'ini epitel hücrelerin, % 16,58'ini lökositlerin, subklinik mastitisli süt örneklerinde İse % 31,44'ünü epitel hücrelerin, % 68,55'ini lökositlerin oluşturduğunu gösterdi. PMN lökositlerin, toplam lökosit sayılarının normal süt örneklerinde % 20,89'unu, subklinik mastitisli süt örneklerinde ise %81,20'sini oluşturduğu saptandı. Sağlıklı ve subklinik mastitisli ineklerden toplanan süt serumlarının mineral madde analiz sonuçları sırasıyla şöyledir; Na, % 43,37±2|92 mg - %58;51±3,68 mg (P<0,005); K, % 97,53±4/44 mg - % 80,37±4,56 mg (P<0;02); Cl, %72,95±3,26 mg - % 108,27±4,50 mg (P<0,001); Pi, %2,34±0/1 *ng - % 2,29±0,04 mg (P>0,90);. Mg, % 2,63±0,07 mg - %2,51±0,04 mg (P>0,20); Ca, % 40,36±2,29 mg - % 49;05±1,85 mg (P<0,05). Sağlıklı ve subklinik mastitisli ineklerden toplanan süt serumlarının metabolit analiz sonuçları sırasıyla şöyledir; glikoz, 3,5210,42 mg/dl - 1,5310,15 mg/dl (P<0;001); kolesterol, 7,2710,76 mg/dl - 8,0410,57 mg/dl (P>0,20); trigliserid, 19,3311,71 mg/dl -25,6411,81 mg/dl (P>0.05). Sonuç olarak, sütte yapılan direkt ve indirekt somatik hücre sayımlarının, süt serumlarında Na, K, Cl gibi mineral maddelerin, LDH, AST, ALT, CK, ALP enzim aktivitelerinin, glikoz metabolitinin saptanmasının subklinik mastitislerin teşhisinde başarılı bir şekilde kullanılabileceği sonucuna varıldı. Ayrıca, eğer uygun koşullar sağlanır ise, subklinik mastitislerin teşhisinde süt serumu örneklerinin AST enzim aktivitesindende faydalanılabilir.
This study was conducted to investigate comparatively various biochemical test on normal and subclinical mastitic milk samples, diagnosed according to the California Mastitis Test (CMT), Direct Microscopic Somatic Cell Count (DMSCC), Differential Cell Count (DCC) results, and to establish usefulness of these tests in the diagnosis of subclinical mastitis cases in cows. In the present study, a total of 100 Holstein, Brown-Swiss cows, in 3-7 years old, supplied by Faculty of Veterinary Medicine University of Uludağ and commercial dairy farms in the region of Bursa, were used as research materials. California Mastitis Test was performed previously on milk samples obtained from all cows. According to the test results, 30 mammary quarters shown (-) reactions were identified as free from subclinical mastitis. The other 70 mammary quarters reacted (?, +,++, +++) were identified as subclinical mastitis. The mean Somatic Cell Count (SCC) were 1.087.571 ±58.489 cells/ml in subclinical mastitic milk samples whereas 134.666±1 5.798 cells/ml in normal milk samples(P<0,001). Differential cell counts performed on normal milk samples indicated that epithelial cells 83,41 % of the total SCC while leucocytes 16,58 % of the total SCC. Subclinical mastitic milk samples 31,44 % epithelial cells and 68,55 % leucocytes. As a result, it was concluded that direct and indirect somatic cell counts of milk, determination of the level of Na, K, Cl.glucose and LDH, AST, ALT, CK, ALP activities in milk serum samples can be used satisfactory! in the diagnosis of subclinical mastitis cases. Furthermore, if appropriate conditions are provided, AST enzyme activity of milk serum samples can be also used in diagnosis of subclinical mastitis cases.
