İntraserebroventriküler verilen CDP-kolin'in serum ghrelin ve leptin düzeyleri üzerine etkisi

Abstract
Ghrelin ve leptin yiyecek alımını kontrol eden iki önemli peptiddir. Yakın zamanda CDP-kolinin iştahı baskılayıcı etkisinin olduğu gösterilmiştir. Bu çalışmada intraserebroventriküler (isv) olarak verilen CDP-kolin?in serum ghrelin, leptin kortikosteron ve glukoz düzeyleri üzerine olası etkilerinin incelenmesi amaçlanmıştır. Çalışmada Wistar-Albino sıçanlara 0.5 (n=8), 1.0 (n=8) ve 2.0 (n=8) µmol CDP-kolin ve tuzlu su isv olarak verildi. Bazal şartlarda ve CDP-kolin sonrası 5., 15., 30., 60. ve 120. dakikalarda serum leptin, ghrelin ölçümü için karotis artere yerleştirilmiş kanülden kan örneği alındı. İkinci aşamada; maksimum etkinliğin görüldüğü doza eşit ekimolar dozda kolin ve sitidin isv verilerek ölçümler tekrarlandı. Üçüncü aşamada; oluşan etkinin muskarinik ya da nikotinik reseptörler aracılığıyla gerçekleştiğinin aydınlatılması açısından CDP-kolin verilmesinden önce atropin ve mekamilamin verilerek deneyler tekrarlandı. 1µmol CDP-kolin enjeksiyonunu takiben serum ghrelin düzeyinde 60. dakikada anlamlı düzeyde baskılanma olduğu (p=0.025), leptin düzeylerinde ise, 60. ve 120 dakikalarda anlamlı artış olduğu (sırasıyla: p=0.012, p=0.017) saptandı. Santral CDP-kolin verilmesinin serum glukoz ve kortikosteron düzeylerinde de zaman ve doz bağımlı artışa neden olduğu belirlendi. CDP-kolin sonrası serum leptin ve ghrelin üzerine olan etkinin benzeri kolin verilmesini takiben gözlenirken, sitidin sonrası herhangi bir etki gözlenmedi. CDP-kolin öncesinde mekamilamin verildiğinde serum ghrelin düzeylerinde görülen baskılanmanın kaybolduğu, leptin artışının ise hem mekamilamin hem de atropin sonrası bloke olduğu belirlendi. Sonuç olarak isv verilen CDP-kolin serum ghrelin düzeylerinde baskılanmaya yol açarken, serum leptin düzeylerini arttırmaktadır Reseptör antagonisti verilmesini takiben gözlenen etkiler ghrelin süpresyonunda nikotinik reseptörlerin, leptin artışında ise hem nikotinik hem de muskarinik reseptörlerin rol oynadığını düşündürmektedir.
Ghrelin and leptin are two important peptides which control food intake. Recently it was shown that CDP-choline has suppressive effect on appetite. In present study we aimed to test intracerebroventricular (icv) administration of CDP-choline on serum ghrelin, leptin, glucose and corticosterone levels in rats. Intracerebroventricular 0.5 (n=8), 1.0 (n=8) and 2.0 (n=8) µmol CDP-choline and saline were administered to male Wistar-Albino rats. For the measurement of serum leptin and ghrelin levels, blood samples were withdrawn baseline and 5th, 15th, 30th, 60th and 120th min after icv CDP-choline injection from a canula that was inserted into the left carotid artery. Equimolar doses of icv choline and cytidine were administered and measurements were repeated at the second stage of the study. Atropine and mecamylamine were injected before CDP-choline and experiments were repeated in the third stage of the study to identify whether the observed effect of the drugs was relevant by muscarinic or nicotinic receptors. After 1µmol CDP-choline injection ghrelin levels were suppressed significantly at 60th min with (p=0.025) whereas serum leptin levels were increased at 60th and 120th min (p=0.012 and p=0.017, respectively). Central CDP-choline injections also induced a dose- and time dependent increase in serum glucose and corticosterone levels. The effect of choline on serum leptin and ghrelin levels was similar by CDP-choline while no effect was seen by cytidine. Suppression of serum ghrelin levels was abolished by mecamylamine pretreatment while increment of leptin was blocked by both atropine and mecamylamine pretreatments. In conclusion; centrally injected CDP-choline was suppressed serum ghrelin levels while increasing serum leptin levels. The observed effects after receptor antagonists suggest that nicotinic receptors play role in suppression of serum ghrelin levels whereas nicotinic and muscarinic receptors both take part in the increment of serum leptin levels.
