Abstract
Bu çalışmada Karacabey Tarım İşletmesi Müdürlüğü At Yetiştirme Şubesinde bulunan safkan Haflinger 4 adet aygır ve 20 adet kısrak kullanıldı. Çiftleşme mevsiminde sun'i vajen yardımıyla alman sperma yağı alınmış kısrak sütüyle sulandırıldı. Sulandırma sonrası ve dondurma sonrası spermatolojik özellikler ile tohumlamalardan elde edilen gebelik oranları incelendi. Aygırlardan toplam 12 ejakülat alındı ve ortalama 20±0.42 mi. hacim, 170.41±2.85xl06/ml. spermatozoon yoğunluğu, %60.83±2.28 motilite, 6.8±0.01 pH, %14.91±0.94 ölü spermatozoa ve %13.16±0.68 anormal spermatozoa saptandı. Sulandırılmış spermayla yapılacak tohumlamalar için sperma 1000 IU/ml. penisilin, 1250 ng/ml. streptomisin içeren yağı alınmış kısrak sütüyle ml.de 50x1 06 motil spermatozoa olacak şekilde sulandırıldı. Sulandırılmış sperma 37°C'de 15 dakika bekletildikten soma spermatolojik özellikleri incelenerek tohumlama uygulamaları gerçekleştirildi. Dondurulmuş spermayla yapılacak tohumlamalar için ise sperma %7 final gliserol içeren aynı sulandırıcıyla 25x1 06 motil spermatozoa olacak şekilde sulandırıldı. Makrotüplerde -125 ile -130°C'de dondurulmuş sperma 45°C'deki su banyosunda 30 saniyede eritildi. Spermatolojik özellikleri incelendi ve tohumlamalar gerçekleştirildi. Sulananına soması %60, eritme sonrası %40 ortalama motilite bulundu. Prostaglandin enjeksiyonundan soma 3-13 gün içinde ovulasyon yapan kısraklara ovulasyon anında veya hemen önce bir kez intrauterin tohumlama yapıldı. On kısrağa 10 mi. sulandırılmış, 10 kısrağa da 4 mi. dondurulmuş sperma ile tohumlamalar yapıldı. Sulandırılmış ve donmuş sperma için sırasıyla %70 ve %50 gebelik oram saptandı.
In the present study, purebred 4 Haflinger stallions and 20 mares, bred in Karacabey Stud Farm Department of Horse Breeding, were used. Semen collected by means of artificial vagina in breeding season was diluted with skimmed mare milk. Spermatological characteristics were examined after dilution and thawing, and conception rates were determined. The averages of volume, spermatozoon concentration, motility, pH, dead spermatozoa and abnormal spermatozoa of total of 12 semen collected from stallions were determined as 20±0.42 ml., 170.41±2.85xl06/ml., 60.83±2.28%, 6.8±0.01, 14.91±0.94% and 13.16±0.68%, respectively. Semen was diluted with skimmed mare milk, cantained 1000 IU/ml. penicillin and 1250 ^g/ml. streptomycin as 50x1 06 motil spermatozoa per msemination dose. Diluted semen stored in water bath at 37°C for 15 minutes, spermatological characteristics were determined and used for inseminations. Semen was diluted with the same diluent contained 7% final glycerol concentration as 25x1 06 spermatozoa per insemination dose for inseminations with frozen semen. Semen, frozen in macrotubes at -125 to -130°C, was thawed in a water bath at 45°C for 30 seconds. Spermatological characteristics were determined and inseminations were performed. Motility averages were 60% and 40% after dilution and thawing, respectively. Single intrauterin inseminations were performed at the ovulation time or just before the ovulation to the mares ovulated 3-13 days after prostaglandin injections. Ten mares were inseminated with 10 ml. diluted semen and 10 mares were inseminated with 4 ml. frozen semen. Conception rates were found 70% and 50% for diluted and frozen semen, respectively.
