Abstract
Bu araştırma, gliserol, DMSO, 1,2 propanediol ve etilen glikol içeren sulandırıcılarla dondurulan koç spermasının değerlendirilmesi ve koyun embriyonik gelişimi üzerine etkilerinin karşılaştırılması amacıyla planlanmıştır. Araştırmada koçlardan sperma elektroejakülasyon yöntemi ile alınmıştır. Sperma alma işlemi aşım mevsimi içinde 5 kez tekrarlanmıştır. Sperma örneklerinin spermatolojik özellikleri incelendikten sonra örnekler bir tüpte toplanmıştır (pooling). Pooling yapılan sperma 1:1 oranında sulandırıcı A ile sulandırıldıktan sonra 4 eşit hacme bölünmüştür ve sulandırıcı B ile 1:1 oranında sulandırılmıştır. Eritme sonrası; motilite, HOST, akrozomal bozukluk (FITC-PSA), akrozomal bozukluk (Gimza) ve DMB oranlarına ait ortalama spermatolojik değerler %6 gliserol içeren grupta sırasıyla 53.3±0.9, 68.3±0.8, 55.3±0.9, 50.5±1.3 ve 3.6±0.3; %6 DMSO içeren grupta sırasıyla 15.3±0.9, 30.4±0.8, 55.6±1.0, 51.7±1.6 ve 3.6±0.2; %6 1,2 propanadiol içeren grupta sırasıyla 40.6±0.8, 52.2±0.8, 49.7±0.9, 44.6±1.3 ve 3.8±0.2; %6 etilen glikol içeren grupta sırasıyla 43.3±0.9, 56.7±0.71, 51.3±0.5, 47.5±0.8 ve 3.9±0.2 olarak tespit edilmiştir. Gruplar arasında motilite, HOST ve akrozomal bozukluk oranları yönünden istatistiksel farklılık önemli bulunmuş (P<0.05), DMB oranlarında ise farklılık bulunamamıştır (P>0.05). %6 Gliserol içeren sulandırıcılarla dondurulmuş koç spermasıyla yapılan IVF'de, 2-4 hücre, 8-16 hücre, morula ve blastosist oranları sırasıyla %79.25, %65.87, %41.27 ve %25.40 bulunmuştur. %6 DMSO grubunda sırasıyla, %38.16, %24.14, %6.90 ve %0 , %6 1,2 propanediol grubunda sırasıyla %72.19, %57.80, %25.69 ve %15.60 ve etilen glikol grubunda ise sırasıyla, %61.94, %56.25, %21.89 ve %11.46 saptanmıştır. %6 Gliserol grubu diğer gruplara göre eritme sonrası motilite ve plazma membran fonksiyonel bütünlüğü daha iyi korumuştur. %40 ve üzerinde motiliteye sahip sperma kullanılarak yapılan IVF sonrası taze spermaya benzer düzeyde bölünme oranı elde edilmiştir.
This study was planned with the aim of evaluating the ram sperm that were cryopreserved with extenders which include glycerol, DMSO, 1,2 propanediol and ethylene glycol and comparing the affects on sheep embriyonic development. Sperm was collected from rams by means of electroejaculator. A total of 5 sperm collection was done during breeding season. After determining the spermatological characteristics of each sperm samples, all samples were collected in one tube (pooled). After pooling, sperm was divided into 4 groups after diluted 1:1 ratio with extender A and diluted 1:1 ratio with extender B. After thawing; general mean rates of motility, HOST, defected acrosome (FITC-PSA), defected acrosome (Giemsa) and DMB in 6% glycerol group were 53.3±0.9, 68.3±0.8, 55.3±0.9, 50.5±1.3 ve 3.6±0.3; in 6% DMSO group were 15.3±0.9, 30.4±0.8, 55.6±1.0, 51.7±1.6 ve 3.6±0.2; in 6% 1,2 propanediol were 40.6±0.8, 52.2±0.8, 49.7±0.9, 44.6±1.3 ve 3.8±0.2; in 6% ethylene glycol group were 43.3±0.9, 56.7±0.71, 51.3±0.5, 47.5±0.8 ve 3.9±0.2 respectively.
