Yeni bir hipotansör drog olan debrisoquine sulfate'ın klinik kullanımı

Abstract

Bu çalışmada, tedavide yeni bir hipotansör drog olarak kullanıma giren debrisoquine sulfate (Declinax) ın hipotansör etkisi ve ayrıca bir Rauwolfia alkotoidi olan Reserpin (Şerpasil) ile hipotansör etki ve yan etkiler bakımından karşılaştırılması yapılmıştır. Topluca 40 vaka üzerinde yapılan araştırmada şu sonuçlar alınmıştır: a) Declinax verilen 30 vakada, diyastolik kan basıncı 2, sistolik kan basıncı 3. haftadan itibaren anlamlı olarak düşme göstermiş ve 1 aylık tedavi sonunda vakaların tümünde kan basıncı kontrol altına alınabilmiştir. b) İlaç çok küçük dozlarda (5-30 mg/ günde) etkili bulunmuştur. c) Yan etki olarak sadece 1 vakada ortostatik hipotansiyon, 2 vakada ekzersiz esnasında gelip geçici baş dönmesi tesbit edilmiştir. d) Serpasil verilen 10 vakada, ortalama kan basıncı değerlerinde ; anlamsız bir düşme görülmüş ve 3 vakada ilaç etkisiz kalmıştır . e) Mutad günlük dozlarda (0, 25-0, 75 mg/ günde) dahi 1 vakada aşırı,2. vakada hafif sedasyon ve psişik depresyon, 1 vakada ishal görülmüştür.

In this study, therapeutic effects of a new antihypertensive drug, debrisoquine sulfate (Declinax) was searched and its hypotensive and side effects was compared with the effects of a Rauwolfia alkaloide (Serpasil). The study was applied to 40 cases and below mentioned results were obtained: a) Declinax caused a significant decrease in the diastolic blood pressure in the second week and also a significant decrease in the systolic pressure during the third week of the trial. Positive effects of the drug on the blood pressure was observed at the end of the fourth week. b) Positive therapeutic effects of the drug was obtained by its very law doses (5- 30 mg/ kg). c) A temperary ortostathic hypotension occured in one case, and a reversible vertigo was observed in two cases during exercises. d) An insignificant decrease was found in the means of the maximal and minimal arteriel blood pressure in the group treated with serpasil. In 3 of the 10 cases of this group no change was observed in the level of blood pressure. e) Therapeutic doses of serpasil (0, 25- 0, 75 mg/ 24 h) caused light sedation in 2 cases and depression in another one.
